Digital PCR Technology

Digital PCR(dPCR) technology is an absolute quantitative nucleic acid detection method, without the need for standard curve to accurately quantify nucleic acid, greatly ensuring the accuracy and precision of detection, is the third generation of quantitative nucleic acid detection technology after real-time fluorescence quantitative PCR technology. It has important application value in liquid biopsy, tumor concomitant diagnosis, noninvasive prenatal screening, pathogen load monitoring and so on.

Microdroplet array printing adopts the original vibration micro(OsciDrop®) core technology to rapidly and stably generate uniform nano-scale microdroplet.

Stable amplification

The nanodroplets are flattened into a single-layer array in the well plate for in-situ amplification and fluorescence reading detection. The operation is convenient and the nanodroplets are effectively used.

Multiple fluorescence

Nanodroplet array reading has high-performance LED excitation light source and orthogonal fluorescence imaging detection system, supports more fluorescence detection channels, and can develop super multiplex digital PCR detection system.

Exact algorithm

Using advanced plan-apochromatic fluorescence imaging correction technology and deep learning nanodroplet array recognition algorithm, the nanodroplet recognition accuracy is high and the signal-to-noise ratio is good.

One-Stop Detection Process
Configure PCR reaction system

Reaction system: 10~25μL.

Nanodroplet generation

Put the reaction system and consumables into the instrument for nanodroplet generation.
Each experiment can generate up to 96 samples; Each print and sample can generate up to 20,000 nanodroplets.

PCR amplification reaction

After the nanodroplets are generated, the instrument seals the amplification area of the well plate to complete the PCR reaction.

Result analysis

Fluorescence photo-detection is performed on each reaction well; the quantitative value of the sample is calculated according to the number of negative and positive nanodroplets, which is according to Poisson distribution.

Application Cases
  • Quantitative detection of COVID-19 Coronavirus N gene and ORF1ab gene

    For the detection of the COVID-19 Coronavirus, Maccura digital PCR is highly sensitive, can quantitatively monitor changes in viral load, and improve the detection rate of weak positive samples. It plays an important role in viral load monitoring, re-examination after discharge and drug efficacy evaluation during the diagnosis and treatment of COVID-19 Coronavirus.

    Nucleic acid standard reference is an important tool for national supervision of nucleic acid testing products. At present, digital PCR is the core method for the research and development of nucleic acid reference materials. As shown in the figure above, among the 9 companies participating in the COVID-19 Coronavirus national reference product calibration of the China National Institutes for Food and Drug Control, the COVID-19 Coronavirus reference calibration results reported based on Maccura digital PCR analysis system are the closest to the COVID-19 Coronavirus national reference product calibration results of the China National Institutes for Food and Drug Control.

  • Absolute quantitative detection of single nucleotide mutation (SNV)

    EGFR gene mutation detection examples: EGFR gene is a receptor for cell proliferation and signal transduction of human epidermal growth factor. This gene is mutated in non-small cell lung cancer. Accurate detection of EGFR gene mutation is of great significance for drug selection in clinical treatment.

  • Absolute quantitative detection of copy number variation (CNV)

    HER2 gene CNV quantitative example: As a proto-oncogene, HER2 gene is overexpressed in breast cancer, lung cancer and other cancers. Accurate quantification of HER2 gene copy number is of great significance for early screening of breast cancer and lung cancer. As shown in the figure above, the representative 2D hash map for the detection of the copy number variation ratio of human HER2 can be used to obtain the CNV rate of HER2 gene by comparing the content of HER2 gene with the internal reference gene.

    24 FFPE tissue samples were verified. The results showed that compared with FISH(in situ fluorescence hybridization) results, the specificity and accuracy of Maccura D 600 automatic digital PCR system were 100%.

  • RNA fusion gene detection

    Periodic monitoring of BCR/ABL expression in patients with chronic myelogenous leukemia (CML) is a prerequisite to ensure the treatment efficacy.
    Digital PCR is more sensitive than RT-qPCR with cellular and peripheral blood samples.
